Photorhabdus ve Xenorhabdus Bakteri Supernatantlarının Farklı Uygulama Şekillerinin Tetranychus urticae Üzerindeki Etkileri

dc.description.abstractEntomopatojen nematodlar (EPN), doğal ekosistemlerde yaygın olarak bulunan ve farklı böcek zararlılarını hedef alabilen, Steinernematidae ve Heterorhabditidae familyalarına ait önemli biyolojik mücadele etmenleridir. Bu nematodlar, simbiyotik olarak taşıdıkları Xenorhabdus ve Photorhabdus türü bakterilerle birlikte konukçuya giriş yaptıktan sonra birkaç gün içerisinde konukçuyu öldürürler. Diğer taraftan tarımsal üretimde önemli ekonomik kayıplara yol açan Tetranychus urticae Koch (Acari: Tetranychidae), geniş konukçu yelpazesi, yüksek üreme hızı ve pestisitlere karşı hızlı direnç geliştirme potansiyeli nedeniyle mücadelesi zor zararlılar arasında yer almaktadır. Bu durum, kimyasal mücadelede karşılaşılan direnç sorunlarını azaltmak amacıyla, alternatif ve çevre dostu mücadele stratejilerine olan ihtiyacı artırmaktadır. Simbiyotik Xenorhabdus ve Photorhabdus bakterileri tarımsal zararlılara karşı biyolojik mücadelede yüksek potansiyele sahip, güçlü ve hızlı etkili sekonder metabolitler ürettiği bilinmektedir. Bu özellikleri sayesinde kırmızı örümceklere karşı kullanılabileceği değerlendirilmektedir. Bu çalışmada, içerisinde sekonder metabolit olduğu düşünülen simbiyotik bakteri supernatantlarının T. urticae üzerindeki doğrudan ve aquaponik sistemde dolaylı olarak etkileri laboratuvar koşullarında değerlendirilmiştir. Uygulamalar kontrol (saf su), besiyeri kontrol ve iki farklı simbiyotik bakteri supernatantı olmak üzere dört gruptan oluşmuştur. Doğrudan etki denemelerinde en yüksek ölüm oranları bakteri supernatantlarında gözlemlenmiştir. Besiyeri kontrol grubu supernatantlara göre daha düşük olmasına karşın saf su kontrol grubundan farklı bulunmuştur. En düşük ölüm oranı ise saf su kontrolünde belirlenmiştir. Aquaponik sistemde yapılan uygulamada ergin başına en yüksek yumurta sayısı kontrol grubunda, en düşük yumurta sayısı ise Xenorhabdus supernatantı uygulamasında belirlenmiştir. Genel olarak bulgular, EPN bakterilerinden elde edilen supernatantların hem doğrudan temasla yüksek ve hızlı öldürücülük sağladığını hem de aquaponik sistemde kök yoluyla uygulandığında T. urticae’nin üremesini azaltabildiğini göstermektedir. Bu sonuçlar, söz konusu metabolitlerin biyolojik mücadelede etkili ve umut vadeden bir biyoakarisit potansiyeline sahip olduğunu düşündürmektedir.
dc.description.abstractEntomopathogenic nematodes (EPNs) are important biological control agents belonging to the families Steinernematidae and Heterorhabditidae, and are widely distributed in natural ecosystems where they can target various insect pests. These nematodes initiate a rapid infection and mortality in the host after entering the body together with their symbiotic bacteria, Xenorhabdus and Photorhabdus. On the other hand, Tetranychus urticae Koch (Acari: Tetranychidae), which causes significant economic losses in agricultural production, is among the most challenging pests to manage due to its broad host range, high reproductive capacity, and strong potential to quickly develop resistance to pesticides. This situation increases the need for alternative and environmentally friendly control strategies to mitigate resistance issues encountered in chemical control. Symbiotic Xenorhabdus and Photorhabdus bacteria are known to produce potent and fast-acting secondary metabolites with high potential for biological control of agricultural pests. Owing to these properties, they are considered promising candidates for use against spider mites. In this study, the direct and indirect (via aquaponic system) effects of symbiotic bacterial supernatants, presumed to contain secondary metabolites, on T. urticae were evaluated under laboratory conditions. The treatments consisted of four groups: control (distilled water), only medium control, and supernatants of two different symbiotic bacteria. In the direct-contact assays, the highest mortality rates were observed in the bacterial supernatant treatments. Although the medium control group exhibited lower mortality compared to the supernatants, it was still significantly different from the distilled water control. The lowest mortality rate was recorded in the distilled water group. In the aquaponic system, the highest number of eggs per female was observed in the control group, while the lowest was found in the Xenorhabdus supernatant treatment. Overall, the findings indicate that supernatants derived from EPN-associated bacteria provide high and rapid mortality through direct exposure and can also reduce the reproduction of T. urticae when applied through plant roots in an aquaponic system. These results suggest that the metabolites in question have effective and promising bioacaricidal potential for biological control.
